The second, called the lysogenic response, results in the covalent integration of the λ genome into the E. coli chromosome where it can remain stably associated as a prophage for many cell generations. Much about these two responses can be explained by what is known about the transcription of the various λ operons.
These heteroduplex molecules contain regions of double-stranded DNA in which the two par ents have homologous sequences and loops or bubbles of single-stranded DNA in which the parental sequences are nonhomologous (see, for exam ple, Figure 9). The lengths of the single- and double-stranded regions can be measured, and by comparing the regions of homology and nonhomology with the genetic properties of the parental DNA's, it is possible to determine the physical positions of the genes on the parent molecules.
